英语翻译The propensity for 1 to bind to DNA and also selectivelyaccumulate within cancer cells in an intact manner promptedus to investigate its uptake and bio-distribution withinindividual A549 human lung carcinoma cells and also itseffect on endogenous elements by means of synchrotron XRF(X-ray fluorescence) imaging methods.Figure 1 shows elementaldistribution maps for a single A549 cancer cell treatedwith 5 mm of 1 for 24 h.The most significant result is that highlevels of both gadolinium and platinum are co-localized withthe intense region in the zinc and phosphorus maps,which isassumed to define the nucleus.These results are consistentwith the cellular uptake of 1 followed by its accumulation inthe nucleus,and the same result was observed in all five cellsmeasured (Supporting Information,Figures S2–S6).The factthat the gadolinium and platinum maps are essentiallyidentical indicates that 1 remains largely intact throughoutthe uptake and accumulation processes,which is consistentwith the results of the ICP-MS experiments (see above).Further support is given by the fact that the XRF-determinedmolar ratio of platinum to gadolinium in the cell matches thestoichiometry of the complex within experimental error (2:1;Supporting Information,Table S3).
